P01 - TRPC1 function in Ca2+ and bone homeostasis
Biochemical identification and functional characterization of TRPC channels in peripheral tissues
TRPC proteins are versatile cellular signal integrators. We will study physiological TRPC1 functions in peripheral tissues (lacrimal gland, bone), extend an in-house made library of antibodies for TRPC proteins (TRPC1-TRPC7) and answer the questions:
1) What is the TRPC protein distribution?
2) Do we work with the correct TRPC cDNAs?
3) Are all TRPC proteins pore-carrying α-subunits?
4) Do β-subunits exist, which bind to the α-subunit and modulate channel function? Parts of these questions will be answered in close collaboration with P02 which pursues a high-resolution proteomic analysis of given TRPC channels in mammalian brain.
