P06 - Role of TPC2 in cellular signaling networks

Two pore channels (TPCs) form a subgroup within the superfamily of TRP channels. Like TRPML1, 2, 3 and TRPM2 these channels are expressed in endo-lysosomes. In this proposal we will focus on the physiological role of TPC2 for vesicle trafficking. In Aim 1, the molecular composition and regulation of TPC2 channel complexes will be studied in HEK 293, in neurons and in hepatocytes using a SILAC based proteomics approach, Ca2+ imaging experiments and lysosomal patch clamp. In Aim 2, the role of TPC2 and proteins interacting with TPC2 for vesicle trafficking, fusion and fission will be analyzed.
