P28 - Role of TPC2 in cellular signaling networks

Two pore channels (TPCs) are endo-lysosomal cation channels which form a subgroup within the superfamily of TRP channels. In aim 1, properties of these channels will be characterized using endo-lysosomal single channel recordings. These experiments will be performed in HEK293 cells, in hepatocytes, cardiac myocytes and hippocampal neurons. In aim 2 the role of TPC2 in atherosclerosis will be analyzed in the ApoE-/- background in mice globally lacking TPC2 channels (TPC2-/-ApoE-/-), and mice with cell type specific and inducible deletion of TPC2 in macrophages, endothelial- or smooth muscle-cells.
